Fat content volume and determining its presence plays a major role in food safety, security, and in nutritional health sector. Tributyrin is one of the triglyceride groups that acts as a liquid fat in the food. In this paper, a study on the functionalized screen-printed electrode (SPE) with graphene (GPH) and gold nanoparticles (GNP) immobilized with lipase is presented. The two-dimensional form of GPH and threedimensional structure of GNP make a suitable surface suitable for the enzyme immobilization. Cyclic voltammetry (CV) measurement of the GPH/GNP/Lipase/SPE has produced a fine reaction. Tributyrin concentration range from 0.1 mg/ml to 1000 mg/ml were measured and detection limit of 0.1mg/ml was achieved. The current response of the GPH/GNP/Lipase/SPE was proportional to the concentration of the tributyrin taken below 1V point at oxidation peak. These results will make the GPH/GNP/Lipase/SPE a quality bioelectrode especially in the field of triglyceride detection.
