基于毒性通路的毒理学危险度评价方法

Abstract

当前的危险度评价(risk assessment)体系主要依赖整体动物试验进行危害鉴定和剂量-反应关系的外推.对于毒效应有阈值的化学物,由动物实验获得未观察到有害作用水平(no observed adverse effect level,NOAEL)、观察到有害作用的最低水平(lowest observed adverse effect level,LOAEL)或基准剂量(benchmark dose,BMD)除以合适的不确定系数得到安全水平或每日允许暴露量;对于毒效应无阈值的化学物,由动物实验获得高剂量的剂量-反应关系,再依靠数学模型分析低剂量暴露的风险,计算危险度。
