Chitosan construction of tissue engineering corneal stroma

Abstract

目的 利用角膜基质细胞与壳聚糖支架共培养构建组织工程角膜基质.方法 将壳聚糖、胶原蛋白、硫酸软骨素、透明质酸钠等按照一定的比例混合,加入原花青素交联剂,构建壳聚糖支架,检测获得的壳聚糖支架的特点及生物相容性.在羊膜上培养兔角膜基质细胞.兔角膜基质细胞与原花青素交联的壳聚糖支架共培养构建初步组织工程角膜基质,行兔角膜板层移植.结果 兔角膜基质细胞可在支架表面上生长,获得初步的组织工程角膜基质.兔角膜板层移植2周后,壳聚糖支架全部脱落.结论 原花青素交联的壳聚糖支架具有良好的生物相容性,在羊膜上培养的兔角膜基质细胞可以保持正常的生物学特性,兔角膜基质细胞和壳聚糖支架可以构建初步的组织工程角膜基质,但不能用于移植。
